McLaren's Lando Norris took the third spot on the podium, making it the first all British podium since 1968.
Seven-time world champion Hamilton last won a grand prix in 2024, when he took victory at the Belgian Grand Prix.
Since signing for Ferrari at the start of 2025, he has struggled to get to grips with the car, but since the new regulations for this season he has been much happier.
He came off the back of two second place finishes at the Monaco Grand Prix and the Canadian Grand Prix, before taking victory in Spain.
It was a race of strategy and Russell was unable to respond to Hamilton and keep the race lead.
Antonelli managed to get the move done on his teammate Russell in the closing stages to take second, but then suffered an apparent power unit issue that took him out of the race.
The gap between Antonelli and Russell in the drivers' championship has closed to 50 points.
Hamilton still sits in second and is only 41 points behind leader Antonelli.
The Brit said after the race on the team radio: "Thank you so much for helping me achieve this dream. I just can't thank you enough. Thank you for everyone pushing so hard at home. So proud of you. To my family, I love you. To the fans, thank you for continuing to remind me who I am. I Couldn't have done it without you."
